Sample preparation

• Soil samples require prior extraction using the sample extraction kit (sold separately)

Sampling time

• Approximately two minutes per sample plus assay run time of approximately 30 minutes

Basic Test Procedure

• Transfer soil or wipe to extraction jar
• Cap and shake for one minute
• Filter extracted sample
• Pipette and dilute samples with dilution ampule
• Transfer 1 ml of buffer to glass buffer tubes
• Pipette and transfer sample to buffer tubes
• Fit antibody coated tubes on buffer tubes; invert and mix;return to antibody coated tube
• Incubate for 10 minutes
• Connect antibody coated and conjugate tubes; invert and mix, return to antibody coated tube
• Incubate five minutes
• Wash antibody coated tubes vigorously four times
• Using a repeater pipette at setting two and 5 mL tip,transfer 200 uL substrateA to each tube
• Add 200 uL substrate B to each tube and mix
• Inclubate for 2.5 minutes
• Add 200 uL STOP to each tube
• Read tubes in photometer.Record data

Basic Test Procedure

• Prepare standards
• Add 100 uL of sample or standard
• Incubate for 30 minutes
• Add 100 uL enzyme conjugate
• Incubate for 30 minutes
• Decant and wash five times
• Add 100 uL colour solution
• Incubate for 30 minutes
• Stop the reaction and read colour at 450nm

Specificity

The EnviroGard® Triazine Test Kit does not differentiate between atrazine and closely related compounds, but detects their presence to differing degrees. The chart below shows the approximate Lower Limits of Detection (LDD) for the cross-reacting compounds.

• Add 160 uL of calibrator, negative control or sample to the tube
• Add four drops of enzyme conjugate
• Incubate for five minutes
• Decant and wash four times
• Add four drops of substrate and chromogen
• Incubate for two minutes
• Add one drop of stop solution
• Read on photometer

• Minimum detection level (MDL) is 0.2 ppm DDT.
• No maximum screening level as extracts can be diluted into test range

• Soil extraction time is typically 2 minutes per sample plus assay run time of approximately 30 minutes.

Specificity

The EnviroGard DDT immunoassay test does not differentiate between DDT and other organochlorines. The table below shows compounds at the minimum detection limit (MDL) -an approximate concentration required to yield a positive result atthe lowest standard. The IC50 is the concentration required to inhibit one half of the color produced by the negative control. It is also used to calculate cross-reactivity values to similar compounds.

Basic Test Procedure

• Extract the soil sample using the MWI Sample Extraction Kit
• Pipet DDT assay diluent into the antibody coated tubes, then add either DDT calibrator or sample extract to the tubes along with DDT enzyme conjugate.
• Wait 15 minutes, then decant and wash the tubes.
• Add substrate.
• Wait for 10 minutes, add stop solution
• Read the absorbance using a photometer, and interpret the results. (Refer to DDT Soil Test Kit Instruction Guide procedures.)
