The QuickChek™ SRB Detection System is a rapid enzyme immunoassay method that detects sulfate reducing bacteria (SRB). The test employs purified antibodies to detect the enzyme adenosine-5'-phosphosulfonate (APS) reductase which is common to all strains of SRB.

The purified antibodies are attached to small particles that selectively capture the APS reductase enzyme. The particles and captured enzyme are then isolated on a porous membrane, which forms a reactive layer that turns blue in the presence of the APS reductase enzyme.

QuickChek™ SRB offers several advantages over cell culture techniques for SRB detection including immediate and accurate results. The kit allows testing of solid and semi-solid samples and detects all SRB, including SRB that is unable to grow in some standard media. The test results are not compromised by chemical or salinity interferences that are often found in field samples.

The QuickChek™ SRB kit is completely self-contained and disposable. Each kit includes all necessary materials to detect and enumerate sulfate reducing bacteria. There is no need for pretreatment or dilution of samples and there are not special hazardous waste disposal limitations.

Sample preparation

• Soil extraction time is typically two minutes per sample plus assay run time of approximately 60 minutes

Specificity

The table below shows compounds at the method detection limit(MDL) which isthe lowest concentration of the compound that can be picked up in the assay. The limit of quantification (LOQ)- an approximate concentration required to yield a positive result at the lowest standard. The IC50 is the concentration required to inhibit one-half of the colour produced by the negative control. It is also used to calculate cross-reactivity values to similar compounds.

Basic Test Procedure

• Add calibrators and sample, enzyme conjugate, and antibody coupled magnetic particles to a test tube. Vortex.
• Incubate for 15 minutes
• Using the RaPID magnetic separator, decant and wash two times
• Add colour solution and incubate for 20 minutes
• Stop the reaction and read colour at 450 nm
• Quantitive results and QC parameters are calculated and printed automatically using the RPA spectrophotometer

EnSys® explosives kits are rapid field or laboratory colourimetric tests for the analysis of explosives in soil. The kits are based on a method developed by Dr. Thomas Jenkins at the US Army Corps of Engineers - CRREL.

Specificity

The EnSys® TNT test is specific for TNT and related explosive compounds and exhibits the following sensitivities. The TNT test does not measure RDX or HMX.

Basic Test Procedure

• Clean cuvettes and set spectrophotometer
• Extract soil sample: Weigh 10 grams of soil sample, Add sample to extraction jar, Measure 50mL acetone into 50mL tube, Pour acetone into extraction jar and shake for three minutes, Allow to settle for five minutes
• Draw 25mL liquid into the syringe from above the sediment layer in extraction jar
• Attach filter tip to syringe and transfer 25mL of sample extract into a cuvette.
• Place cuvette in spectrophotometer and record initial absorbance
• Add one drop of developer solution, cap and shake for three seconds.
• Remove stopper and place the'sample' cuvette in cell holder and read sample absorbance.
• NOTE: For analysis of samples containing DNT,and/or where DNT concentration is of concern, samples must be allowed to develop for 10 minutes before reading sample absorbance. This will not affect colour development for other nitroaromatics.
• Record data and calculate results.
